Under the supervision of Christophe BRUEL and Mathias CHOQUER
Titled Of content, space and time: characterization of secretory vesicles in the phytopathogen Botrytis cinerea, the aim of the thesis was to characterize the protein content (Axis 1) and spatial and temporal dynamics (Axis 2) of secretory vesicles in Botrytis cinerea. In Axis 1, we developed protocoles for the enrichment of secretory vesicles that led to the obtention of, to our knowledge, the first proteome of secretory vesicles in a filamentous fungus. Axis 2 led to the development of image and signal analysis pipelines in Python to study spatial and temporal dynamics from confocal time-lapse acquisitions. This study led to the identification of subpopulations of hyphae regarding the dynamics of secretory vesicles accumulation in growing filaments.
